Melanotan II was developed by Hruby & Hadley at the University of Arizona. Phase I clinical trials by Dorr et al. (Clinical Pharmacology & Therapeutics, 1996) demonstrated dose-dependent melanogenesis and tanning in fair-skinned subjects. The sexual arousal side effects led to development of PT-141 (bremelanotide). Safety concerns include potential interactions with melanocytic nevi, and case reports have described changes in existing moles. Barnetson et al. (2006) studied MT-II as UV photoprotection in fair-skinned individuals, showing increased melanin density. The peptide has not progressed to Phase III trials due to the broad receptor activation profile and associated side effects. Category 2 classification reflects FDA concerns about non-selective melanocortin activation.
