REV-ERB Agonist (Circadian/Metabolic Modulator)
Also known as: SR9011, SR-9011, SR 9011
CAS 1379686-29-9Formula C23H31ClN4O3SPubChem CID 57394021
SR-9011 is a synthetic small-molecule REV-ERB (Rev-erbα/β) agonist — not a peptide — studied preclinically as a circadian and metabolic modulator. It has no human data, no regulatory approval, and is WADA-prohibited at all times. It is commonly confused with "Stenabolic," which is properly its analog SR9009, not SR-9011.
SR-9011 is an agonist of REV-ERBα and REV-ERBβ. Binding enhances REV-ERB-mediated transcriptional repression (recruiting the NCoR corepressor), repressing core clock genes such as Bmal1 and downstream metabolic genes in liver, muscle, and adipose (Solt et al., Nature 2012). All data are cellular or rodent.
In diet-induced obese mice (intraperitoneal dosing), SR-9011/SR9009 increased energy expenditure, reduced fat mass, and improved dyslipidemia and hyperglycemia (Solt et al., Nature 2012). Human liver microsome work identified 14 metabolites but no half-life, and the authors caution against human extrapolation. There are no registered human trials of SR-9011; no human safety, dose, or pharmacokinetic data exist.
