Gonadorelin (GnRH pulse), Kisspeptin-10 (upstream GnRH trigger), and Enclomiphene (estrogen receptor modulation) target the hypothalamic-pituitary-gonadal axis at three different levels. Often used in PCT (post-cycle therapy) research contexts or for studying endogenous testosterone restoration.
Gonadorelin is a synthetic form of gonadotropin-releasing hormone (GnRH) used to maintain fertility and endogenous testosterone production during TRT. It is commonly co-prescribed with testosterone replacement therapy to prevent testicular atrophy and preserve HPG axis function.
Role: GnRH agonist. Directly pulses the pituitary to release LH and FSH. The standard research tool for maintaining gonadotropin signaling.
Kisspeptin-10 is a truncated fragment of the kisspeptin neuropeptide that plays a central role in reproductive hormone signaling. It is a potent stimulator of GnRH release and is studied for its role in fertility, puberty, and hypogonadism.
Role: Upstream GnRH trigger. Activates kisspeptin receptors on GnRH neurons in the hypothalamus, stimulating endogenous GnRH pulsatility.
Enclomiphene is the trans-isomer of clomiphene citrate, a selective estrogen receptor modulator (SERM) that stimulates endogenous testosterone production. While not a peptide, it is widely tracked in the peptide and TRT community as an alternative or adjunct to testosterone replacement therapy.
Role: Selective estrogen receptor modulator (SERM). Blocks hypothalamic estrogen feedback, disinhibiting GnRH/LH secretion. Oral, non-peptide compound commonly paired with peptide protocols.
